Correlation of Yeast Measurement Between Spin-Down and Coulter Counter Laboratory Analysis and In-Line Measurements With the McNab Cell Counter

Abstract
This work addresses the use of an in-line, across the pipe-measuring analyzer to measure yeast cell count in brewery applications. Data is presented showing the correlation between the in-line McNab cell counter and laboratory analysis including: ASBC spin-down 0-65% and Coulter Counter measurements. Cell concentration levels at various stages of the brewing process are ultra-high; with concentrations as high as 2 billion cells/ml being typical in such areas as the pitching line and at the bottom of fermentation vessels. The McNab HSA4 in-line cell counter eliminates common sampling and dilution errors often found in the laboratory, and replaces the necessity and expense of tedious laboratory analysis. Optical adjustments have improved the accuracy of the cell counter from 70% to 98% as compared to lab methods, making this analyzer suitable for controlling feed-forward and feed-back control loops involving cell cropping, pitch blending and transfer of yeast. The HSA4 uses a near-infrared radiation signal and is designed to be mounted directly in the pipe line typically 2 to 6 inches (50 - 150mm) in diameter. This cell counter allows the brewer to better control yeast content so as to meet specifications, improve uniformity of fermentations, and enhance filtering operations.

Este trabajo se refiereal uso de un analizador en linea, atravez de la tuberia para medir la cuentade celulas de levadura en aplicaciones de cerveceria. Los datos son presentadosmostrando la correlacion entre el aparato de conteo de celulas en lineaMcNab y analisis de laboratorio, incluyendo: mediciones ASBC de centrifugaciono-65% y mediciones de conteo de culturas. Los niveles de concentraciones en varias etapas del proceso cerveceroson ultra altos; con concentraciones desde 2 mil millones de celulas pormililitro, siendo estas tipicas en areas como en la linea de inyeccion oen la salida de los fementadores. El contador de celulas en linea McNab HSA4 elimina errores comunes demuestreo y dilucion frecuentes en el laboratorio, y reemplaza la necesidady costo de analisis laborioros en laboratorio. Ajustes opticos han mejoradola exactitud del contador de celulas de 70% a 98% comparado con metodosde laboratorio, convirtiendo a este analizador en apto para controlar circuitosde entrada y salida que involucren transferencia de levadura, inyeccion,etc. El HSA4 usa una se�al de radiacion casi infrarojo y esta dise�adopara ser montado directamente en la tuberia, tipicamente de 2 a 6 pulgadas(50-150MM) de diametro. Este contador de celulas permite al cervecero unmejor control del contenido de levadura para satisfacer mejor las especificaciones,mejorar uniformidad de las fermentaciones, y mejorar las operaciones defiltracion.
