​106. Resonance light scattering technique for the determination of proteinase A activity

ABSTRACT: A simple and sensitive method had been developed for the determination of proteinase A activity using a resonance light scattering (RLS) technique. The method was based on the principle that TCA and trace protein can form association particles and the particles’ concentration proportioned resonance scattering light intensity under certain conditions. In this study, several methods were compared for proteinase A activity measurement. In the meantime, the influence of temperature, pH value, and TCA concentration were investigated. This new method was fast and convenient and had a low detection limit (0.595 g/mL) and wide linear range (1–200 g/mL). The system had better tolerance against interfering substances, and the reaction system remained stable for at least 150 min. The method was satisfactorily applied to the determination of trace proteinase A activity.
