"Identification of Microbial Populations Associated with Sorghum Fermentation," by E. D. Deenanath, S. E. Iyuke, and D. Lindsay, has been published in the MBAA
Technical Quarterly.
Abstract
This study reports on the isolation and identification of microbiological populations that are unique to the University of the Witwatersrand microbrewery plant during sorghum fermentation. For the fermentation procedure, the hydrolysate was propagated with either
Saccharomyces cerevisiae strain NRRL Y2084 or
Issatchenkia orientalis. Microbiological procedures such as plate counts and PCR were performed at the postmashing and postfermentation stages to detect the dominant microorganisms during sorghum processing when inoculated with starter cultures. Microbial isolates were characterized by sequencing and phylogenetic analysis. At the postmashing stage, no microbial contaminants were found to be associated with the process. At the postfermentation stage, plate counts showed microbial counts between 5.00 and 8.00 log CFU/mL. The characterization of microbes isolated at the postfermentation stage was based on PCR amplification of the 16S and internal transcribed spacer (ITS) regions. Sequencing results identified the isolates as
Lactococcus lactis, Lactococcus garvieae, Lactobacillus casei, Enterococcus faecalis, Saccharomyces cerevisiae strains NCL117 and T8,
Saccharomyces paradoxus, Saccharomyces pastorianus, Saccharomyces kudriavzevii, Issatchenkia orientalis, and
Candida inconspicua.
"The Enzymology of Cell Wall Breakdown During Malting and Mashing: An Overview," by C. W. Bamforth, has been published in the MBAA Technical Quarterly.
Abstract
Enzymolysis of the cell walls of the starchy endosperm of barley can be considered a two-stage process: solubilization and polymer digestion. Solubilase enzymes include not only carboxypeptidase acting as an esterase, but also other enzymes, notably an early-developing xylanase. It is supposed that pentosan is primarily located on the outside of the cell wall. Nonetheless, pentosan is less efficiently degraded than
b-glucan, probably because the chief xylanases are blocked by an endogenous inhibitor. Although there can be very efficient depolymerization of
b-glucan, it appears that degradation is incomplete and that the primary hydrolysis products are
b-linked oligosaccharides. This is likely because the exo-glucanases that hydrolyze them develop late in germination and also have a very low affinity for their substrates. The advantage of this for the brewer is that beer can be legitimately claimed as a good source of putative prebiotics.

The 2010 MBAA Brewery Packaging Technology Course is in full swing, with a full-house of 50 attendees. We toured the New Glarus brewery this past Saturday, and brewmaster and brewery owner Dan Carey hosted one of the tour groups. Attendee Jim Maliziola (MillerCoors) led one of the group discussions.

The discussion will be on methods of using the media to establish and grow direct customer relationships for small brewers.

District St Louis to Hold Meeting at Sage Bistro |
|
MBAA District St. Louis will hold its regular monthly meeting this evening, Thursday, March 18, at Sage Bistro in the shadow of the Anheuser-Busch St. Louis brewery. Rob Naylor (Anheuser-Busch) will be confirmed as our new District secretary, a position recently vacated due to a resignation. The dinner presentation will be given by Gordon Gosejohan (DSI Process Systems) on the history and manufacturing techniques of sanitary process fittings and equipment used in the food, beverage, and pharmaceutical industries.
